Effects of inhibitors of the toll-like receptor-4 (TLR4) signaling cascade. Trophoblast cells were cultured with 500 µM palmitic acid (PA) for 24 h after adding inhibitors of TLR4 signaling (100 mM CLi-095 or 200 µg/ml anti-TLR4 antibody) and nuclear factor kappa Beta (NFkB) signaling pathways (5 µg/µl anti p65) for 1 h. Interleukin-6 (IL-6), interleukin-8 (IL-8) and tumor necrosis factor-alpha (TNF-α) mRNA were measured by RT–PCR and data were normalized for beta actin. Results are shown as means ± SEM, n = 14, and 1.5 × 106/well primary cultured cells were plated in 12-well plate for each condition, *P < 0.05.
