Enzymatic activity of caspases against keratin-17. Radioactive-labeled keratin-17 was translated using the TNT Quick Coupled Transcription/Translation System in the presence of [³⁵S]-methionine. Cleavage reactions were performed with or without recombinant caspase-3 or -7 as described in Materials and methods. Samples were separated by 12% SDS–PAGE. [³⁵S]-labeled proteins were detected by autoradiography. The upper band indicates intact [³⁵S] keratin-17 and the lower band designates cleaved [³⁵S] keratin-17.
