Effects of USP33 knock-down on CP110 protein levels and anaphase catastrophe in human lung cancer cells. (A) A549 and (B) Hop62 cells were independently transfected with each of two different USP33 targeting siRNAs or control siRNA. Forty-eight hours after USP33 knockdown, cells were treated with seliciclib (10μM) for 24 hours and fixed and scored for multipolar anaphases. Expression of CP110 protein at each time point was shown using immunoblot analyses and ImageJ quantification. Experiments were independently replicated at least three times. Quantifications by ImageJ were done by combining for analyses all independent experimental results.
