Single cysteine mutants retain cholesterol regulation.
A–E, CHO-7 cells were transfected for 24 h with the indicated single cysteine mutants. Cells were then pretreated overnight with compactin (5 μm) and mevalonate (50 μm) and then treated for 8 h with Chol·CD (20 μg of cholesterol/ml), compactin, and mevalonate. Protein was harvested, separated by 10% SDS-PAGE, and then immunoblotted with anti-V5 using GAPDH as a loading control. F, densitometry from A–E.
