Effects of dexamethasone on TNF-α-induced CXCL1 secretion and signaling. (A) HUVECs (left) and human A549 epithelial cells (right) were treated with TNF-α in the absence or presence of dexamethasone (Dex) for 6 h. The CXCL1 concentration in the culture media was analyzed by ELISA. (B) HUVECs were pretreated with Dex (500 nmol/L) for 30 min, followed by stimulation with TNF-α (5 ng/mL) for (a) the indicated time intervals or (b) 15 min. The cells were collected and analyzed by RT-PCR or Western blotting (WB). The PCR products of MKP-1 and β-actin were 219 and 314 bp, respectively (n=3−4). bP<0.05, cP<0.01 vs TNF-α alone.
