Model depicting the proposed mechanism by which NGF-activated MAPK signaling affects TRPV2 to enhance neurite outgrowth. Target-derived NGF binds to TrkA at the end of neurites. The complex is internalized into signaling endosomes and transported toward the nucleus in the cell body. At the nucleus, ERK signaling can increase TRPV2 expression. Within endosomes in the neurite shaft, TrkA activates ERK, which can phosphorylate TRPV2. Changes in Ca2+ due to increased TRPV2 protein expression and activity then enhance neurite outgrowth.
