Tregs stimulation of ESC proliferation and invasiveness is dependent on IL-10 and TGF-β. (a) We treated ESCs isolated from eutopic endometrium from patients with endometriosis with rhIL-10 and/or rhTGF-β for 48 h and analyzed the proliferation (a), invasion (b) and the expression of MMP2, TIMP1 and Cox-2 (c). In addition, ESCs were co-cultured with CD4+CD25+ Tregs and/or anti-human IL-10, TGF-β, CD28, CTLA-4, CD39, CD73 neutralizing antibodies for 48 h. BrdU proliferation (d), Matrigel invasion (e) and In-cell Western assays (f) were performed to examine the proliferation, invasiveness, and MMP2, TIMP1 and Cox-2 expression in ESCs, respectively.
