The interaction between PRR14 and HP1α is critical for C2C12 myogenesis. (a) Mutant PRR14-overexpressing C2C12 stable cell line was established and its expression was confirmed by immunoblotting. The cells were induced for myogenesis in DM for 5 days and representative phase-contrast images are shown (b), the percentage of cells with multiple nuclei (>4 N) was quantified by FACS and statistically analyzed (c). The expression of MHC was detected by immunostaining (d). And the statistical analysis of apoptotic cells from FACS data is presented in e. All FACS data are presented as mean±S.D. (n=3) and statistically analyzed by two-tailed student's t-test. (f) Again, cells were subjected to TUNEL assay and counterstained with DAPI. To quantify the percentage of TUNEL-positive cells, cells were counted in three different fields and 200 cells were counted for each field. The data are presented as mean±S.D.
