EP₄ and PKA are involved in PGE₂-induced mTORC1 activation. A: PANC-1 cells were treated with the EP₄ agonist CAY10580 (10 μM) for 15 min. B: PANC-1 cells were pretreated with the EP₄ antagonist ONO-AE3-208 (1 μM) for 1 h and then stimulated with or without PGE₂ for 15 min. C: PANC-1 cells were pretreated with the PKA inhibitor H-89 (5 μM) for 1 h and stimulated with or without PGE₂ (1 μM) for 15 min. After treatments, cell lysates were collected and subjected to immunoblotting with indicated antibodies. GAPDH was used as a loading control.
