(A) Total anti-HER-2 specific serum IgG induced upon vaccination with 4TO7 PMVs protein transferred with GPI-HER-2. HER-2 expression was similar in all protein transferred samples at 0.034 μg HER-2/μg PMV. Serum was collected from groups of vaccinated mice (n = 5) day 7 post boost. Flow cytometry analysis using 1:200 diluted serum incubated with D2F2/E2, D2F2, or 4TO7 cells was performed. (B) IgG subtype analysis of anti-HER-2 antibodies in sera from vaccinated mice. 1:100 diluted serum collected on day 7 post boost from vaccinated mice (n = 5) was incubated with 50, 000 D2F2/E2 cells per well in triplicate in a cell ELISA. The subtype of bound antibody was detected by adding peroxidase conjugated subtype specific antibodies.
