Proposed model for regulation of telomeric silencing by sumoylation. Sir2 is sumoylated by Siz2 and in unperturbed cells both unmodified and sumoylated Sir2 coexist. The fraction that is sumoylated at K215 is unable to interact with Sir4 and hence is not recruited to telomeres. Conditions that promote sumoylation, especially at K215, would shift the equilibrium towards decreased telomeric silencing.
