Ageing impairs CNS remyelination, partially through impaired phagocytosis of myelin debris, which contains inhibitors of oligodendrocyte progenitor cell differentiation. Natrajan et al. identify retinoid X receptor (RXR) signalling as a genetic and pharmacological target to reverse this impairment, and successfully enhance myelin phagocytosis in monocytes from patients with multiple sclerosis.
