SIGIRR is required for LPS tolerance in T24 and 5637 cells. LPS tolerance was induced in SIGIRR and negative control siRNA cells by pretreatment with 100 ng/ml LPS (1st LPS) for 24 h or maintained in medium. After 24 h, the cells were washed with fresh medium and restimulated with 100 ng/ml LPS (2nd LPS) for 6 h. The levels of IL-6 and IL-8 in the supernatants were measured by ELISA. Experiment were repeated at least three times in triplicate with similar results, #p < 0.05 with respect to the same treatment but without pretreatment with LPS, *p < 0.05 versus negative control siRNA with the same treatment
