IL-1β treatment decreases SERCA2b protein stability in INS-1 cells and isolated rat islets. INS-1 cells (ac) or isolated rat islets (de) were treated with 1 μM actinomycin or 10 μM CHX combined with or without 5 ng/ml of IL-1β for indicated times. Total RNA and protein were isolated, and RNA was subjected to quantitative real-time PCR (qRT-PCR) for quantification of SERCA2b and actin transcript levels. Immunoblot was performed using antibodies against SERCA2 and actin. Protein and mRNA levels were plotted relative to levels at time zero, and one-phase decay lines for each treatment are shown. Indicated comparisons are significantly different (*P<0.05 and ***P<0.001)
