A and B. After 7 days, bone marrow cells from integrin β3−/− have limited attachment to extracellular matrices. C. ScaI and c-Kit positive CACs were isolated from bone marrow of wild type mice and the purity was confirmed. The purified cells from bone marrow and integrin β3−/− mice were seeded (~1 ×104) on 48-well plates coated with varied extracellular cellular matrices. After 7 days culture, cell numbers were counted. D and E. Bone marrow cells from wild type and integrin β3−/− mice were labeled with different antibodies and subjected to flow cytometry. Monocyte (CD45), macrophage (CD11b), neutrophil (Gr-1) populations and Sca I/c-Kit double positive CACs were not different between the two types of mice. F and G. The CACs from lineage negative population in bone marrow cells were isolated from wild type and integrin β3−/− mice (F). The lineage negative population was gated for isolation of Sca I+/c-Kit+ CACs and the adhesion analysis was performed (G).
