Polarized light microscopy of lamellar bodies. (A) Single ATII cell imaged by polarized light microscopy. LBs appear as bright, birefringent structures with a central extinction cross (inset), whereas other cellular structures remain invisible. (B) The increase in temperature leads to a disappearance of the polarization signal. (C) Plot is showing the inverse correlation between polarization signal and temperature. (D) Plot is demonstrating the correlation between temperature-dependent polarization intensity and the temperature-dependent GPF of Laurdan. Data shown are means ± standard deviation from in total 60 single LBs and six independent experiments.
