Modulation of total brain-derived neurotrophic factor (BDNF) and BDNF-6 expression by EZH2. (A) Knockdown of EZH2 in N2A cells reduced expression of EZH2 (left panel). Knockdown of EZH2 induced up-regulation of total BDNF mRNA 24 and 48 hours after the nucleofection, and up-regulation of BDNF-6 after 48 hours (central and right panels). Expression levels were normalized to β-actin and Gapdh and results were expressed as mean ± standard error of the mean (SEM; n = 5 independent biological replicates). *p < 0.05; **p < 0.01 compared to scramble siRNA (SCR) by Student’s t-test. (B) mRNA levels of total BDNF and BDNF-6 in N2A cells were increased after 24 hours of treatment with 3-deazaneplanocin A (DZNep) at 0.5, 1, and 2 μM.
