(A) Fancb−/y mice show BM failure after MMC treatment. Fancb−/y mice and their male WT littermates were injected with 0.3 mg/kg MMC. Hematological parameters were analyzed 8 weeks after MMC injection. Results are means ± standard deviation (SD) of three independent experiments (n = 9–12 per group). (B) MMC decreases BM cellularity of Fancb−/y mice. Total BM mononuclear cells from mice described in (A) were isolated and enumerated. (C) MMC treatment reduces SLAM and LSK cell number in Fancb−/y mice. The frequencies of SLAM and LSK cells in WT and Fancb−/y mice treated with or without MMC were analyzed by flow cytometry. (D) MMC treatment leads to the death of Fancb−/y mice. Fanca−/−, Fancb−/y mice and their male WT littermates were injected with a single dose of MMC (1 mg/kg) and survival of the animals was monitored by using the Kaplan-Meier curve method and analyzed by the log-rank test.
