Effect of IL-2 or IL-15 pretreatment on LTB4-induced NK cell migration. (a) PBLs were incubated for 18 h in the absence or presence of IL-2 (50 ng/mL) or IL-15 (10 ng/mL). Cells were stained with anti-CD3 and anti-CD56 antibodies and then placed in a chemotaxis assay in response to LTB4 10−9 M or 10−6 M. Migrating CD3 CD56+ cells were counted by FACS, and results were expressed as migration index (LTB4/vehicle). Bar graphs represent mean ± SEM of seven independent experiments. P < 0.05 by paired Student's t-test, treated versus untreated cells. (b) IL-15-activated PBLs were preincubated with or without U75302 or LY255283 before chemotaxis assays.
