Quantification of flow cytometry shows depletion of neutrophils after CFA injection and plantar incision. (A) By using i.p. injections of the Gr1 antibody, and its isotype (IgG) as a control, CD11b+Ly6G+ neutrophils were depleted from the inflamed hind paw 24 h after intraplantar injection of CFA. The infiltration of all three Ly6C populations of CD11b+Ly6G− monocytes/Mø were unaltered at this same time point (P ≥ 0.272; n = 5–7 per group). (B) CD11b+Ly6G+ neutrophils were depleted from the inflamed hind paw 24 h after incision following i.p. injections of the Gr1 antibody, relative to its isotype (IgG) control. The infiltration of Ly6Clow and Ly6Chi myeloid cells was unaltered, whereas there was a modest, albeit significant, reduction in Ly6Cmed cells (n = 3 per group; *P < 0.05, t test).
