Analysis of short-term HBV-specific functional T cells in HLA-A2/DR1 AAV-HBV-persistent mice following a single TG1050 immunisation. HLA-A2/HLA-DR1 transgenic mice were intravenously injected with PBS or AAV-HBV (day 0) and immunised 1 month later (day 32) with either TG1050 or empty adenovirus. Induced HBV-specific immune responses were assessed 2 weeks after TG1050 or empty Ad immunisation through (A) IFNγ ELISPOT assay on spleen cells and interferon γ (IFNγ)/tumour necrosis factor α (TNFα) intracellular cytokine staining assays using (B) spleen cells and (C) liver-infiltrating lymphocytes. The analytical conditions are as described in figure 3, except that HLA-A2-restricted peptides were used (see online supplementary table S1). Data from two experiments are presented. In experiment 2, data from the group PBS+TG1050 could not be exploited due to high background. NE, non-evaluable.
