Aspirin induces interleukin-4 (IL-4) mRNA expression and protein production. (a) HMC-1, EoL-1 and Jurkat cells were treated with various concentrations of aspirin for 4 h. IL-4 expression was measured by quantitative real-time PCR (qPCR); n=4–6. The IL-4 mRNA levels (means±s.e.m.) are expressed relative to the PP1A expression levels (**P<0.01 compared with unstimulated cells at 4 h). (b) Kinetics of IL-4 expression in the presence of 5 mM aspirin; n=2. (c) Cells were transfected with an IL-4 promoter reporter (−649). After 24 h, the transfected cells were stimulated with aspirin for 4 h; n=4 (*P<0.05 and **P<0.01 compared with unstimulated cells at 4 h). (d) Cells were treated with aspirin for 24 h, and the levels of intracellular IL-4 were analyzed by FACS (fluorescence activated cell sorting).
