Figure 6. Effects of saracatinib plus cetuximab combination on tumor growth, survival and signal transduction of mice xenografted with H1975 erlotinib-resistant tumors.

Figure 6

A. After 7 days following subcutaneous injection of H1975 cells, mice were randomized (10/group) to receive cetuximab, saracatinib or their combination, as described in the Methods section. The one-way ANOVA test was used to compare tumor sizes among treatment groups at the median survival time of the control group (38.5 days). The results are statistically significant for the combination versus control or saracatinib (P < 0.0001). Bars, SDs. B. Median survival differences were statistically significant for the saracatinib versus control groups (log-rank test); mice treated with cetuximab or with the combination did not reach a median survival, since 70% and 80% of animals were still alive at the end of the experiment, respectively. C. Western blot analysis was performed on total lysates from tumor specimens of two mice sacrificed on day 14. Tumors derived from each treatment group were pooled during lysis to obtain a single specimen.
