(A) Relative mucin mRNA expression for the hTCEpi cell cultures at day 0 (unstratified cells) and at days 1 to 6 during the stratification process. For unstratified cells, the expression of MUC1 was low, and the expression levels of MUC4 and MUC16 were negligible. Expression of mucin mRNA reached a peak between day 1 and day 3 (n = 3). (B) Western blot analysis showed expression of the mucin protein for hTCEpi cell cultures at day 0 (unstratified cells) and at days 1 to 6 during the stratification process. For unstratified cells, expression of all mucins is negligible. Expression levels of the proteins MUC1 and MUC16 reproduced the observed mRNA expression profile, whereas expression of the MUC4 protein suggested a monotonic increase during the stratification process.
