Antitumor efficacy of chloroquine (CQ) in vivo. (A and B) CQ significantly inhibited tumor growth as determined by fluorescence imaging. Nude mice with HepG2-GFP orthotopic human liver cancer were administered CQ according to Materials and methods. (A) Tumor size was monitored twice a week by fluorescence imaging. Scale bar, 1 µm. (B) The data were converted to tumor growth curves. (C and D) CQ treatment significantly reduced tumor volume. Mice were sacrificed on the 25th day after treatment (the end of study, n=7). Tumor tissues were harvested, photographed (C) and weighed (D). (E) No obvious toxicity against body weight gain was observed during treatment. The body weight of animals was measured twice a week during the treatment period. (F) CQ treatment decreased the expression of Ki-67 and enhanced the expression of cleaved PARP in vivo. Tumor tissues were collected; paraffin-embedded. The expression of Ki-67 and cleaved PARP was detected using immunochemistry. Scale bar, 20 cm.
