Plasmid constructs used in pull-down assays. The plasmids are derivatives of pYW15b (Wang et al., 2000). The VirA receiver domain (RA) is constitutively expressed from PN25 and includes a FLAG tag at the carboxyl end (pAW184). The VirG DNA-binding domain (DNABV G) and the TorR DNA-binding domain (DNABTR) are constitutively expressed from PN25 (pAW187 and pAW192, respectively). Each construct carries an RGS-6XHis tag near the amino terminus. The molecular weight, including epitope tags, is indicated for each protein fragment.
