Massive cytokine secretion after electroporation with mRNA encoding a DC-activating protein. DCs were electroporated either without RNA (Mock) or with RNA encoding the tumor antigen MelanA (MelanA) or MelanA and constitutively active stabilized mutated IKKβ. Twenty-four hours after electroporation, supernatants of the cells were taken and the cytokine secretion by the cells was determined in a cytometric bead array. The cytokine concentrations in the supernatants are depicted. Each symbol represents an individual donor, tested in an independent experiment (n = 8).
