Impact of injection with Tc24 on IgM-mediated hydrolysis. Balb/c mice were injected with either 25 μg Tc24 + 200 μg alum or 200 μg alum only (control). (A) IgM (50 μg/mL) from the Tc24 injected group and from the alum injected group was incubated with 2.6 μM Tc24 for 40 hours at 37°C with shaking, and then subjected to sodium dodecyl sulfate polyacrylamide gel electrophoresis using 15% Tris-Gly gels. Percent cleavage was calculated using ImageJ. Representative silver-stained 15% Tris-Gly gels of Tc24 hydrolysis by IgM purified from either (B) 25 μg Tc24 + 200 μg alum injected mice or (C) alum-only injected mice. Numbers indicate individual IgM samples purified from mice in respective groups tested in Tc24 hydrolysis assays, in duplicate.
