FHDC1-induced Golgi dispersion is unaffected in centrinone-treated cells. NIH 3T3 cells were treated with the Plk4 inhibitor centrinone (300 nM) for 14 d, and the effects of this treatment on FHDC1-induced Golgi dispersion were assessed by immunofluorescence. (A) mCherryFP-tagged FHDC1 (red) was expressed by transient transfection in control cells that were subsequently stained with anti-giantin (white) and anti–γ-tubulin (green) antibodies. (B) mCherryFP-tagged FHDC1 (red) was expressed by transient transfection in centrinone-treated cells that were subsequently stained with anti-giantin (white) and anti–γ-tubulin (green) antibodies. FHDC1 expression is sufficient to induce Golgi dispersion in both cases, even though the centrosome can no longer be detected in centrinone-treated cells. Scale bar, 10 μm.
