Virulence plasmid-dependent inhibition of IL-8 secretion by PMNs incubated with the enteropathogenic Yersinia species. Y. enterocolitica, (Y. ent.) and Y. pseudotuberculosis (Y. pstb.) strains, with (pYV+) or without (pYV−) their native virulence plasmid were incubated with human PMNs and the amount of IL-8 secreted was compared to PMNs incubated with pCD1+ (KIM5) or pCD1− (KIM6) Y. pestis. Y. pestis engineered to express yopP instead of yopJ (yopJ-yopP) was also incubated with PMNs. Culture supernatant was collected after 12 h of incubation and the concentration of IL-8 was determined by ELISA. For these experiments the bacteria were not opsonized before addition to the PMNs. The mean and SEM for at least three independent experiments, except for the yopJ-yopP strain, for which only two experiments were realized, are indicated.
