Mice were injected with AAV-IL-22 (3×107TU/mouse), AAV mock (3×107TU/mouse) or normal saline three days before the first 2-OA-OVA immunization and sacrificed at week 10. (A) The frequency of regulatory T cells (CD3+CD4+Foxp3+) in the liver was measured. Liver mononuclear cells were stained with anti-CD3, CD4 and Foxp3 Abs and assayed by a flow cytometer. (B) The expression of IL-10 in the liver was measured by qRT-PCR. This data represents the fold increase of data normalized to the naïve mice. n=15 mice per group. *, p<0.05; **, p<0.01. (C) Liver mononuclear cells from 2-OA-OVA immunized mice were collected and IL-22R expression was measured by RT-PCR.
