Kinetics of growth and plaque morphology in cell culture. Different cell lines or primary cultures were infected with A12-WT or P1-deoptimized (P1deopt) FMDV at an MOI of 10. After 1 h of incubation, unabsorbed virus was removed by washing with 150 mM NaCl–20 mM MES (pH 6.0), followed by addition of complete MEM. Infected cells were frozen at 1, 3, 6, and 24 hpi. Titers of viruses released after thawing were determined by plaque assay on BHK-21 cells.
