Association of MV DI RNA with PACT. (A) HEK293 cells stably expressing FLAG-PACT were transfected with MV DI RNA. After 6 h and before harvest, cells were treated with formaldehyde. Cell lysates were immunoprecipitated with anti-FLAG antibody, and total RNA was extracted from the precipitates. PACT and α-tubulin in the input lysate (5%) were visualized by Western blotting with anti-FLAG and anti-α-tubulin antibodies, respectively. DI RNA was detected by agarose gel electrophoresis after RT-PCR with DI RNA-specific primers. (B) SLAM-expressing A549 cells were infected with the Hu-191 virus at a multiplicity of infection of 0.5. After 18 h, cells were treated with formaldehyde and cell lysates were immunoprecipitated (IP) with anti-PACT antibodies. The input lysate (5%) was also probed for PACT and α-tubulin. The experiments were repeated twice, and similar results were obtained each time.
