The function of heterochromatin protein 1γ (HP1γ) in the DNA damage response pathway. (A) Bloom syndrome protein (BLM) and proliferating cell nuclear antigen (PCNA) were associated with endogenous HP1γ proteins. BLM and PCNA were coimmunoprecipitated with anti-HP1γ antibodies using HEK293T cell lysates and immunoblotted with indicated antibodies. (B) HeLa cells were treated with 50 μM of etoposide or vehicle (dimethyl sulfoxide, DMSO) for 6 hours and immunostained with the indicated antibodies. (C) HeLa cells were transfected with pSUPER-siHP1γ and immunostained with the indicated antibodies. (D) Nonproportional Venn diagram showing subsets of HP1γ-interacting proteins functionally related to DNA damage response pathways.
