The specificity of HIV-1 RNA detection in living cells by using TIRF microscopy. Constructs 1-Gag-BSL and 1-GagCeFP-BSL were cotransfected into human cells with Bgl-YFP (A) or MS2-mKate (B), followed by TIRF microscopy. Gag signals were detected in the CeFP channel whereas RNA signals were detected in the YFP channel (A) or the mKate channel (B). Similarly, 1-Gag-MSL and 1-GagCeFP-MSL were cotransfected with MS2-mKate (C) or Bgl-YFP (D); RNA signals were detected in mKate channel (C) or YFP channel (D).
