(B) Fluorescent microscopy confirms that GFP is robustly expressed in the heart of Myh6-Cas9-2A-GFP transgenic animals. (Left) A negative littermate control. (C) Myh6-Cas9-2A-TdTomato animals display strong expression of TdTomato in the heart by fluorescent microscopy, whereas a negative littermate control (Left) had no TdTomato expression. (D) Western blot confirms that 3x-Flag–tagged Cas9 is correctly produced in both Myh6-Cas9-2A-GFP and Myh6-Cas9-2A-TdTomato lines.
