EMSA reveals T₃-induced dissociation of MBP-NCoR1–540 from the THRβ1·FAM-hPL promoter complex, whereas Fe³⁺-heme causes dissociation of THRβ1 from DNA. A, binding assays were prepared in 0.5× TNGD buffer containing 0.25 mg ml⁻¹ BSA, 25 ng μl⁻¹ poly(dI-dC), 1 μm FAM-hPL promoter, and the indicated concentrations of THRβ1, MBP, MBP-NCoR1–540, and T₃. B, same experimental setup as described for A except containing no BSA or T₃ and with the indicated concentrations of Fe³⁺-heme. Assays were incubated on ice for 2 h prior to separation by native gradient gel electrophoresis and imaging of the protein-DNA complexes.
