Inhibition of dynamin GTPase activity decreases p-ERK activity in multiple gonadotrope cell lines. A, αT3–1 cells were pretreated (30 min) with vehicle (0), dynasore (80μM), or dyngo (30μM) for 30 minutes followed by a 10-minute treatment of either 0 or GnRHa (10nM). Cells were lysed in RIPA buffer and Western blotted using an anti-p-ERK1/2 antibody. After probing with p-ERK, membranes were stripped and reprobed with an antibody that detects β-tubulin to assess protein lane loading. All groups were normalized to β-tubulin and are represented as fold change relative to 0. B, LβT2 cells were treated as described in A. All groups compared with GnRH: *, P < .05.
