Downregulation of miR-375 confers DDP resistance upon SGC7901 human gastric cancer cells. (A) SGC7901 cells transfected with a miR-375 inhibitor or miR-control were treated with DDP for 48 h, after which cell viability was determined using the Cell Counting kit-8 assay. SGC7901 cells transfected with the miR-375 inhibitor had a significantly increased survival rate, as compared with the miR-control and SGC7901 parental cells. (B) FACS analysis demonstrated that downregulation of miR-375 expression levels significantly reduced the sensitivity of SGC7901 cells to DDP-induced apoptosis following 48 h of treatment with 0.5 µg/ml DDP, as compared with the miR-control and SGC7901 parental cells.
