Overexpression of ErbB2 in breast cancer cells increases mitochondrial localization and activity of HK II. (A) Overexpression of ErbB2 upregulated the expression of HK II. Whole cell lysates were collected and for western blotting analysis, and β-actin was used as a loading control. (B) Overexpression of ErbB2 did not alter the expression of HK II in cytoplasm but increased the distribution of HK II in mitochondria. The cytosolic and mitochondrial fractions of cells were isolated. Prohibitin was used as a mitochondria marker and loading control. α-tubulin was used as a cytoplasm marker and loading control. (C) The activity of HK was measured in the breast cancer cell lines. The results were normalized to the amount of total protein compared with the control cells.
