(A) RAW264.7 cells were treated with LPS (100 ng/ml) + IFN-γ(20 ng/ml) for 24 h as negative control, and treated with IL-4 (20 ng/ml) + IL-13 (20 ng/ml) +0, 4, 8 mg/ml Huaier extract for 24 h. The relative mRNA expression of M2 macrophages markers including Arg-1, CD206, IL-10 and MRC-2 was examined using qPCR. (B) Immunofluorescence of CD206 after treatment with LPS + IFN-γ, IL-4 + IL-13, IL-4 + IL-13 + 4 mg/ml, or IL-4 + IL-13 + 8 mg/ml Huaier extract for 24 h. (C) FITC-Dextran phagocytosis assay of RAW264.7 cells after treated with Huaier extract for 24 h. Cells treated with LPS + IFN-γ was used as a positive control. (D) Western blotting of Arg-1 and iNOS protein expression. (E) Relative Arg-1 and iNOS protein expression level. Results are presented as the mean ± SEM of triplicate samples. *P < 0.05, **P < 0.01.
