(A) The induction by doxycycline of AMOTL1 expression in BC52 tet on AMOTL1 for 48 hours (Dox +, in red) has no visible impact on endogenous Yap (green) distribution when compared with uninduced cell (Dox −). Yap is essentially excluded from the nuclei of the cells in colonies. (B) Extracts were prepared under the same growth conditions as in A. The induction of AMOTL1 expression shows no noticeable impact on endogenous Yap expression or phosphorylation on S127. (C): IHC staining of YAP in BC52 tumors of mice fed with (DOX +) or without (DOX −) doxycycline in the drinking water. Nuclear and cytoplasmic expression of YAP appears essentially unaffected by AMOTL1 overexpression.
