Both Gβγ and Ras regulation of PI3Kγ are crucial for accumulation of PtdIns(3,4,5)P3 at the leading, up-gradient edge. Polar plots of neutrophils, responding to an fMLP-containing micropipette, which expressed eGFP-PH-PKB in different genetic backgrounds (x wild type (WT); x p110γ−/−; x p110γDASAA/DASAA; x p101−/−) or pre-treated with a p110γ inhibitor (AS252424; 10 μM, using x WT cells).
