A–E. A549 cells were transduced with lentivirus encoding empty vector or SDC-1 cCTF. Transduced A549 cells were analyzed for proliferation measured as changes in density over 48 h (A) Cells were grown to confluence on collagen G (B) or fibronectin (C) coated wells and wounded by a defined scratch. Wound closure was monitored continuously for 24 h and quantified as percent wound closure in relation to full wound closure using the IncuCyte ZOOM. D–E) Transduced cells were wounded by scratching and subsequently covered with matrigel to analyze cell invasion from the wound edges into the matrigel within the wounded area. Results were expressed as percent of invaded cells in relation to the empty vector control (D). Exemplary images of three independent invasion experiments are shown and the injured area at time point 0 h is indicated by white lines (E). All data were calculated as means + SD from three independent experiments. Statistically significant differences compared to empty vector are indicated by asterisks (p < 0.05).
