A–F. A549 cells were transduced with SDC-1 (A–C) or scr shRNA (D–F) Subsequently, a second transduction was carried out with virus for overexpression of SDC-1 cCTF or empty vector control. Efficiency of overexpression was controlled by quantitative PCR with primers annealing to the C-terminus of SDC-1 allowing to detect both endogenous SDC-1 and overexpressed cCTF mRNAs and expressed in relation to GAPDH mRNA (A and D). A549 cells expressing SDC-1 cCTF or empty vector were analyzed for changes in density over 48 h (B and E). Double transduced A549 cells were grown to confluence on collagen G coated wells and wounded by a defined scratch. Wound closure was monitored for 24 h and quantified using the IncuCyte ZOOM (C and F). All data were calculated as means + SD from three independent experiments. Statistically significant differences compared to corresponding empty vector control are indicated by asterisks (p < 0.05).
