Chimeric Antigen Receptor (CAR) Design. CARs consist of an extracellular antigen-binding domain, a hinge region, a trans-membrane domain and an intracellular signaling domain. First generation CARs include only CD3ζ as their signaling domain. Second generation CARs include a co-stimulatory domain derived from CD28 or 4-1BB.
