HMW-AF extract induced the expression and activation of tyrosine-protein phosphatase nonreceptor type 11 (PTPN11). (A) Submerged NHBE cells were stimulated with 1:320 wt/vol HMW-AF extract for 24 hours, and cell lysates were collected to analyze the expression of PTPN2, PTPN6, PTPN10, and PTPN11 mRNA by RT-PCR. (B) NHBE cells were treated with HMW-AF for the indicated time points, and cellular protein was collected for Western blot. (C) Expressed bands for phospho-PTPN11 were quantified using Image J software and normalized to internal control actin.
