RAW264.7 cells were stimulated with 500 ng/ml visfatin for different times (0, 2, 4, 8 and 12 h). Protein was extracted, respectively, from the cytosol and nucleus and the expression levels of p65 and IκBα were, respectively, detected. A and B, Western blot analysis was used to detect the expression of p65 in the nucleus. C and D, Western blot analysis was used to detect the expression of p65 and IκBα in the cytosol. E and F, The NF-κB pathway was blocked with 2 h of treatment with the following inhibitors: BAY11-7082 (20 μM) and SC-514 (20 μM) before visfatin was added to the cells. Western blot analysis was used to detect the expression of MMP-8. ** P<0.01 versus the control group, ## P<0.01 versus the visfatin group. Data shown are the means ± SEM from three independent experiments that were performed in duplicate.
