Figure 2

IL-1β reduces JNK1 but not JNK2 or JNK3 protein expression and increases P-JNK after 0.5 h of exposure in INS-1 cells. (a) INS-1 cells were exposed to 150 pg/mL IL-1β or vehicle for 4 to 24 h. JNK subtypes protein expression, JNK1 (circle), JNK2 (square), and JNK3 (triangle), were measured by immunoblotting and normalized to actin or tubulin. Data are shown as % IL-1β-regulated JNK subtype expression of their respective nonexposed controls. Data are shown as means of n = 3–5 independent experiments ± SEM; P < 0.05 versus 4 h time point. (b) INS-1 cells were exposed to 150 pg/mL IL-1β or vehicle for 0.5 to 24 h. INS-1 cells exposed for IL-1β for 12 h were lysed at an earlier time point paralleled with the 12 h vehicle ctrl. P-JNK was measured by immunoblotting and normalized to T-JNK. Data are shown as means of n = 4 independent experiments ± SEM; P < 0.05, ∗∗ P < 0.01, and ∗∗∗ P < 0.001 versus vehicle (0 h). Representative gels are shown.
