Figure 7. FST inhibits SiO2 NP-induced ROS production.

Figure 7

(A) A549 cells transfected with siRNA targeting FST (siFST) or FST gene (FST/OE) were treated with or without 50 μg/ml of SiO2 NPs for 12 h. Intracellular ROS generation was detected by DCFH-DA fluorescent probe and flow cytometry. (B) Relative ROS level was calculated. The values are the mean ± SD of three independent experiments. **P < 0.01. (C) A549 cells transfected with siRNA targeting FST (siFST) were treated with or without SiO2 NPs for 12 h. Mitochondrial ROS was detected using MitoSOX. (D,E) A549 cells transfected with control siRNA or siRNA targeting FST (siFST) were treated with or without 50 μg/ml of SiO2 NPs for 12 h. The mRNA level of NOX1 (D) and NOX5 (E) was measured by real time qPCR. (F,G) A549 cells overexpressing FST gene were treated with or without 50 μg/ml of SiO2 NPs for 12 h. The mRNA level of NOX1 (F) and NOX5 (G) was measured by real time qPCR. The data are presented as the mean ± SD of three independent experiments. **P < 0.01.
